ABSTRACT

This study aimed to compare the predictive value of six selected anthropometric indicators for benign prostatic hyperplasia (BPH). Males over 50 years of age who underwent health examinations at the Health Management Center of the Second Xiangya Hospital, Central South University (Changsha, China) from June to December 2020 were enrolled in this study. The characteristic data were collected, including basic anthropometric indices, lipid parameters, six anthropometric indicators, prostate-specific antigen, and total prostate volume. The odds ratios (ORs) with 95% confidence intervals (95% CIs) for all anthropometric parameters and BPH were calculated using binary logistic regression. To assess the diagnostic capability of each indicator for BPH and identify the appropriate cutoff values, receiver operating characteristic (ROC) curves and the related areas under the curves (AUCs) were utilized. All six indicators had diagnostic value for BPH (all P ≤ 0.001). The visceral adiposity index (VAI; AUC: 0.797, 95% CI: 0.759-0.834) had the highest AUC and therefore the highest diagnostic value. This was followed by the cardiometabolic index (CMI; AUC: 0.792, 95% CI: 0.753-0.831), lipid accumulation product (LAP; AUC: 0.766, 95% CI: 0.723-0.809), waist-to-hip ratio (WHR; AUC: 0.660, 95% CI: 0.609-0.712), waist-to-height ratio (WHtR; AUC: 0.639, 95% CI: 0.587-0.691), and body mass index (BMI; AUC: 0.592, 95% CI: 0.540-0.643). The sensitivity of CMI was the highest (92.1%), and WHtR had the highest specificity of 94.1%. CMI consistently showed the highest OR in the binary logistic regression analysis. BMI, WHtR, WHR, VAI, CMI, and LAP all influence the occurrence of BPH in middle-aged and older men (all P ≤ 0.001), and CMI is the best predictor of BPH.

ABSTRACT

In recent years, the field of network pharmacology (NP) has developed rapidly, but the flawed and routine workflow has seriously affected the scientificity and reliability of NP analysis results. For complex diseases caused by environmental and genetic factors, symptomatic treatment or drugs targeting a single pathophysiological process cannot prevent or delay the progression of the disease, so the drug development fails or withdraws from the market. Therefore, there is an urgent need to develop new ideas for NP analysis that combines multiple pathophysiological processes. The key pathophysiological process is an important and complete set of pathological changes in the process of the occurrence, development, and outcome of the disease, which represents the current comprehensive and profound understanding of the nature of the disease. In order to improve the quality of NP research and promote the healthy development of the NP field, this paper proposes a new idea of NP analysis based on key pathophysiological processes. Based on the long-term clinical practice of traditional Chinese medicine and the key pathophysiological process of the disease, the method comprehensively analyzes the pharmacological mechanism and active ingredients of traditional Chinese medicine compound from the perspective of key pathophysiological process, which increases the scientifically, reliability, and repeatability of the analysis results. This paper takes Alzheimer's disease (AD) as an example to illustrate the necessity, feasibility, main workflow, advantages, and disadvantages of this method, and it is expected to screen disease-modifying drugs that prevent or reverse the course of the disease and promote the clinical transformation of research results.

ABSTRACT

Network pharmacology (NP) is a novel interdisciplinary subject based on the combination of systems biology, multi-omics theory, computer synthesize, network database, and so on. It is widely used in traditional Chinese medicine (TCM) for active component screening, compatibility rule, pharmacological mechanism, and toxicity-efficacy network. Domestic NP-related papers began to boost from 2017, but some research showed abnormal "homogenization" in the screening of key components. Due to the non-standard and unreasonable situation in early NP analysis, the screening results always contained the same and widely-existed 200-500 Chinese medicine substances as the key components for TCM compounds, regardless of the various “disease-treatment-medicine” approaches to the study. If the "homogenization" phenomenon cannot be promptly clarified and corrected, it will lead to the misunderstanding that the components such quercetin, kaempferol, and sitosterol are "guaranteed to cure all diseases", which overestimates the pharmacological weights of the components in each disease. This phenomenon seriously interferes the selection and quality control of Q-Markers related to TCM compounds. It violates and negates the scientific connotation of "treating the same disease with different therapies" and "treating different diseases with the same therapy" guided by the holistic view and the theory of syndrome differentiation and treatment. In the long term, the "homogenization" phenomenon may even hinder the healthy development of NP in TCM. Based on TCM theory and modern medicine, this paper started with the "thomogenization" of component screening in NP, and analyzed from the three module of "tphenomenon consequences", "tcause exploration", and "tsolving strategies", thus providing important references for NP research methods.

ABSTRACT

OBJECTIVE@#To investigate the therapeutic mechanism of resveratrol (RES) for Alzheimer's disease (AD) in light of network pharmacology.@*METHODS@#We searched PubChem, BATMAN-TCM, Genecards, AD, TTD, String 11.0, AlzData, SwissTargetPrediction, Metascape and other databases for the therapeutic targets of RES and human AD-related targets. The intersection was determined using Venny 2.1 to obtain the therapeutic targets of RES for AD. The protein-protein interaction (PPI) network was constructed, the gene ontology (GO) was enriched and the Kyoto Encyclopedia of Genes and Genomes pathway (KEGG pathway) were analyzed. Cytoscape 3.7.1 software was used to construct a target-signaling pathway network of RES in the treatment of AD. Molecular docking verification was carried out on SwissDock (http://www.swissdock.ch/docking). We examined a 293Tau cell model of AD for changes in protein levels of pS396, pS199, Tau5, CDK5, glycogen synthase kinase 3β (GSK3β) and p-GSK3β in response to RES treatment using Western blotting.@*RESULTS@#We obtained 182 targets of RES, 525 targets related to AD, and 36 targets of RES for AD treatment, among which 34.6% of the targets were protein-modifying enzymes, 27.7% were metabolite invertase, 13.8% were gene-specific transcriptional regulators, and 10.3% were transporters. The core key targets of RES in the treatment of AD included INS, APP, ESR1, MMP9, IGF1R, CACNA1C, MAPT (microtubule- associated protein Tau), MMP2, TGFB1 and GSK3B. Enrichment analysis of GO biological process suggested that the biological function of RES in AD treatment mainly involved the response to β-amyloid protein, positive regulation of transferase activity, the transmembrane receptor protein tyrosine kinase signaling pathway, regulation of behavior, learning or memory, aging, and transmembrane transport. KEGG pathway enrichment analysis showed that the most significantly enriched signaling pathways were AD pathway, PI3K-AKT signaling pathway, cGMP-PKG signaling pathway, and MAPK signaling pathway. Molecular docking results showed that RES had strong binding with ESR1, GSK3B, MMP9, IGF1R, APP and INS. In the cell model of AD, treatment with 50 μmol/L RES for 12 h significantly reduced the levels of pS396 and pS199 by regulating CDK5 and GSK3β activity (@*CONCLUSIONS@#RES produces therapeutic effects on AD by acting on multiple targets and affecting multiple signaling pathways and improves AD-associated pathologies

ABSTRACT

Purpose@#Circular RNAs (circRNAs) are a new class of RNA molecules whose function is largely unknown. There is a growing evidence that circRNAs play an important regulatory role in the progression of a variety of human cancers. However, the exact roles and the mechanisms of circRNAs in gastric cancer are not clear. In this study, we aimed to elucidate the mechanism of hsa_circ_0005556. @*Materials and Methods@#Real-time quantitative polymerase chain reaction was used to detect the expression of hsa_circ_0005556, miR-4270, and matrix metalloproteinase-19 (MMP19) in gastric cancer tissues and cell lines. The expression of hsa_circ_0005556 in gastric cancer cells was silenced by lentivirus, and cell proliferation, invasion, migration, and tumorigenesis in nude mice were assessed to evaluate the function of hsa_circ_0005556 in gastric cancer. @*Results@#The expression of hsa_circ_0005556 in gastric cancer tissues and gastric cancer cell lines was higher compared to normal controls. In vitro, the downregulation of hsa_ circ_0005556 significantly inhibited proliferation, migration, and invasion of gastric cancer cells. In vivo, the downregulation of hsa_circ_0005556 suppressed tumor growth in nude mice. @*Conclusions@#Our study shows that the hsa_circ_0005556/miR-4270/MMP19 axis is involved in proliferation, migration, and invasion of gastric cancer cells through the competing endogenous RNA (ceRNA) mechanism.

ABSTRACT

Objective To observe the multimodal imaging characteristics of combined hamatoma of the retina and retinal pigment epithelium (CHRRPE).Methods A retrospective case study. From January 2013 to December 2017,6 CHRRPE patients (6 eyes) diagnosed in Department of Ophthalmology,Sun Yat-sen Memorial Hospital were included in the study. There were 4 males and 2 females, with the mean age of 12.0±8.10 years. There were 5 eyes with BCVA ≤0.1,1 eye with BCVA> 1.0.Corneal fluoroscopy showed 1 eye with an external oblique 15°,and the remaining eye had no abnormalities in the anterior segment. All eyes underwent fundus color photography,FAF,FFA,ICGA,OCT and color Doppler flow imaging (CDFI).The multimodal imaging characteristics were observed. Results All the affected eyes CHRRPE were located in the posterior pole and showed mild elevation. Most of the retinal neuroepithelial layers had different degrees of hyperplasia, vascular tortuosity and retinal folds.Of the 6 eyes,4 eyes (66.7%) involving the macula and optic disc, only 2 eyes (33.3%) involving the macula.OCT showed that the structure of the neuroepithelial layer was unclear and the signal intensity was uneven;it involved 2 eyes of the whole retina (33.3%) and only 4 eyes of the neuroepithelial layer (66.7%).FFA and ICGA showed that the choroidal background fluorescence of the early lesions was weakened, and the lesions showed slightly weak fluorescence;the late telangiectasia fluorescein was obviously leaked, and the lesions were stained with fluorescence.FAF mainly appears as weak autofluorescence with a small amount of strong autofluorescence.CDFI has no characteristic performance. Conclusions CHRRPE is mainly a membrane-like hyperplasia without angiogenesis, involving the retinal neuroepithelial layer, and may also involve the entire retina.OCT is dominated by strong reflection;AF,FFA and ICGA are mainly weak fluorescence.

ABSTRACT

·AIM: To comprehensively investigate the relationship between outer retinal layer thickness and age in normal eyes. ·METHODS: One hundred normal eyes of 100 subjects who underwent spectral - domain optical coherence tomography ( SD - OCT ) were included in this retrospective study. The distances between the external limiting membrane ( ELM ) line and the photoreceptor inner segment/outer segment ( IS/OS ) line ( ELM-IS/OS), the IS/OS line and the cone outer segment tips (COST) line ( IS/OS-COST), the COST line and the retinal pigment epithelium ( RPE) complex ( COST-RPE) and the full retinal thickness ( RT) were measured at the fovea and on four quarters. The relationship between thickness and age or sex was then analysed. ·RESULTS: A thinner RT was observed in women in a multiple regression analysis ( men: 234. 47 ± 16. 79 μ m;women: 223. 13±15. 43 μ m). The RT on the nasal quarter and the ELM-IS/OS thickness at the fovea and on the four quarters were significantly and negatively correlated with age. The IS/OS-COST and COST-RPE thicknesses at the fovea and on the four quarters were not significantly correlated with age or sex, respectively. The RT at the fovea was significantly thinner than on the four quarters. The ELM - IS/OS, IS/OS - COST and COST - RPE thicknesses at the fovea were significantly thicker than on the four quarters. ·CONCLUSION: In normal eyes, the RT thickness on the nasal quarter and the ELM - IS/OS thickness were significantly and negatively correlated with age. The IS/OS - COST and COST - RPE thicknesses were not significantly correlated with age or sex.

ABSTRACT

Objective By comparing the methylation status of imprinted gene H19 in three different quality spermatozoa and the methylation status of the embryos after the combination of spermatozoa, the sperm methylation status in the ART technique was preliminarily explored with the postnatal embryo methylation. Methods In this study, a total of 91 spermatozoa and 91 low-quality embryos were collected from IVF-ET in the First People's Hospital of Yunnan Province. Among them, sperm samples were divided into three groups according to the sperm concentration parameter in the WHO fourth edition, then the relationship between different quality spermatozoa and postnatal embryo methylation were analyzed. Results The abnormality rate of H19 methylation status in group B was significantly higher than that of the sperm group and the embryo group. Conclusions The abnormalities of H19 methylation mainly concentrated in the abnormal parameters of spermatozoa, suggesting that the abnormal state was related to the decrease of sperm quality;Through IVF, ICSI fertilized embryos have the same proportion of the corresponding abnormal state with no significant difference; Abnormal sperm embryos did not show abnormality, which may be related to the self-repair function of the embryo itself.

ABSTRACT

Bardet-Biedl syndrome (BBS) is a rare genetic disease caused by ciliary structure abnormality or dysfunction. To date, more than 21 BBS genes (BBS1 - 21) have been reported to independently cause the disorder. Although the cellular functions of BBS proteins are not yet fully understood, model organisms have revealed that such proteins are involved in ciliary functions and intracellular transport. Among the 21 BBS genes, BBS7 is unique in that its product is a subunit of the BBSome and can directly interact with the BBS chaperonin complex. Previous studies using animal models showed that BBS7 mutation can cause similar phenotypes to human patients, and human disease caused by BBS7 variants are special and more complex. This article reviewed recent progresses on BBS7.

ABSTRACT

<p><b>OBJECTIVES</b>To investigate the impacts of laparoscopic bariatric surgery on fasting glucagon-like peptide-1 (GLP-1) and Ghrelin level in patients with type 2 diabetes mellitus (T2DM), and the mechanism in surgical treatment of T2DM.</p><p><b>METHODS</b>From March 2010 to August 2011, 44 patients with T2DM underwent laparoscopic bariatric, including laparoscopic Roux-en-Y gastric bypass (LRYGB, n = 14), laparoscopic mini-gastric bypass (LMGB, n = 11), laparoscopic sleeve gastrectomy (LSG, n = 9) and laparoscopic adjustable gastric banding (LAGB, n = 10). The curative effects, changes of metabolism and gastrointestinal hormones were analyzed respectively.</p><p><b>RESULTS</b>Within 6 months after surgery, the clinical complete remission of T2DM was 11, 8, 6, 3 cases in LRYGB, LMGB, LSG, LAGB group respectively; the clinical partial remission was 3, 3, 2, 4 cases respectively. The inefficacy was 1, 3 patients in LSG and LAGB group respectively. The effects of surgery within 6 months postoperative among 4 groups were different (χ(2) = 8.162, P < 0.05). The levels of body mass index (F = 275.29) and homeostasis model assessment of insulin resistance (F = 40.09) of 4 groups were declined in 6 months postoperatively (P < 0.01). The extents of decrease were no significance among 4 groups. Compared to preoperative level, GLP-1 in LRYGB ((116 ± 33) vs. (66 ± 20) ng/L and LMGB group ((103 ± 22) vs. (65 ± 16) ng/L) was higher in the first month after surgery (F = 21.76 and 139.21, P < 0.05). The changes in LSG and LAGB group were no significance (P > 0.05). The level of Ghrelin in LRYGB, LMGB, LSG group at the first week after surgery were (208 ± 79), (275 ± 102) and (258 ± 91) ng/L respectively, and they were lower than preoperative (there were (398 ± 114), (439 ± 96) and (446 ± 105) ng/L, F = 55.08, 49.96 and 46.47, all P < 0.01). But the level of Ghrelin in LRYGB and LMGB groups rebounded in the first postoperative month. The postoperative level of Ghrelin was higher in LAGB group (F = 29.24, P = 0.001).</p><p><b>CONCLUSIONS</b>There are difference efficacies and impacts on gastrointestinal hormones among different modes of bariatric surgery. The change of gastrointestinal hormones is plausible mechanism of T2DM remission after surgery.</p>

Their structures were identified on the basis of physicochemical property and multiple spectral data.</p><p><b>RESULT</b>Nineteen phenolic compounds were separated from 95% ethanol extracts from P. tricholobus Franch. and identified as sulfonated benzyl glucoside (1), 3-(4beta-D-glucopyranosyloxy-3, 5-dimethoxy) -phenyl-2E-propenol (2), dihydrosyringin (3), tangshenosides II (4), 4-hydroxy-2,6-dimethoxyphenol-1-O-beta-D-glucopyranoside (5), 4-hydroxymethyl-2, 6-dimethoxyphenyl-1-O-beta-D-glucopyranoside (6), arbutin (7), rutin (8), kaempferol-3-O-alpha-L-rhamnopyranosyl-(1 --> 6)-beta-D-glucopyranoside (9), quercetin-3-O-beta-D-glucopyranoside (10), kaempferol-3-O-beta-D-glucopyranoside (11), afzelin (12), petasiphenol (13), caffeic acid (14), chlorogenic acid (15), 2-hydroxy-5-acetylbenzoic acid (16), p-hydroxy-benzoic acid (17), protocatechuic aldehyde (18) , and p-hydroxy-phenylpropionic acid (19).</p><p><b>CONCLUSION</b>Above result shows that phenolic compounds contained in P. tricholobus mainly include simple phenols, phenolic glycosides, coffee acid and flavonoid glycosides. Among them, compound 1 was separated from the composite family for the first time; compounds 2-7, 9, 11, 12, 16, 19 were separated from the genus Petasites for the first time, and the others were separated from the plant for the first time. These compounds have been proved to have pharmacological effects such as anti-inflammation, antibiosis, antioxidantion.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To observe the effect of Gegen Qinlian Decoction (GQD) in combination with short-term intensive insulin treatment on type 2 diabetes mellitus (T2DM) of dampness-heat syndrome and its influence on dosage of insulin used.</p><p><b>METHODS</b>The GQD group (n = 14) was treated by GQD and insulin, while the conventional group (n = 16) was given insulin intensive treatment alone.</p><p><b>RESULTS</b>In the GQD group, the treatment was markedly effective in 5 patients, effective in 6 and ineffective in 3, the total effective rate being 78.6%, much better than that in the conventional group (2, 7, 7 and 56.3% respectively, u = 2.58, P < 0.01). And it took less time for controlling blood glucose (BG) in the GQD group (4.54 +/- 0.50 days) than that in the conventional group (5.31 +/- 0.57 days, P <0.01); furthermore, by the end of the treatment course, as compared with that at the time just after BG being controlled, the daily average insulin dosage used in the GQD group reduced by 9.07 +/- 6.51 U, while it was only 4.38 +/- 5.94 U in the conventional group, showing significant difference between them (P < 0.05).</p><p><b>CONCLUSION</b>Based on short-term insulin intensive treatment, the combined using of GQD could reduce the dosage of insulin used and shows better clinical curative effect for patients with T2DM of dampness-heat syndrome.</p>
